Targeting Inflammasome Activation To Combat Infections With Intracellular Pathogens (Old ID 20303)

The project will investigate the potential to specifically target intracellular sensors, so called inflammasomes, to enhance immunity against the intracellular pathogens Mycobacterium tuberculosis and Toxoplasma gondii. Particular emphasis will be placed on the regulation of inflammasome-mediated secretion of iterleukin-18 and subsequent initiation of protective interferon-gamma production. The project aims to identify novel host-pathogen interactions that can enhance effective control and clearance of these pathogens and to test the associated molecules in murine infection models as well as primary human samples. Results obtained from this research project may aid in the development of novel vaccine candidates and immunomodulatory strategies against intracellular pathogens.

The QoVAX SET (Queensland COVID-19 Vaccination Safety and Efficacy Trial) program is a population-based study of COVID-19 vaccine safety and effectiveness, providing a resource for studies to understand how the immune system responds to the COVID-19 vaccines, and what factors might affect vaccine response. Though this contract, JCU is supporting the North Queensland arm of this program - QoVAX Program participants recruited in Cairns.

Our overall working hypotheses are: 1. The rational design of tropical disease vaccines and other therapeutics requires improved understanding of immunology, host-parasite interactions and application of innovative bioengineering. 2.Improved interventions will be required to eliminate tropical infectious diseases. Our Specific Aims are: 1. Discover pathogenic and immune mechanisms, protective antigens, biomarkers and therapeutic targets for the control of tropical infectious diseases. 2. Develop new vaccines, therapeutics and diagnostics. 3. Optimise human pathogen challenge models to study host-pathogen interactions and test new vaccines and therapeutics. 4. Develop and test new clinical and public health interventions in disease-endemic settings.

The research team will undertake collaborative research in rural and remote communities of SA and QLD which will contribute directly to the prevention of chronic disease at various stages along the care continuum. They will investigate community and health service based strategies such as obesity prevention, nutrition improvement to prevent the onset of chronic diseases such as diabetes, renal and cardio vascular disease in health adults and children. The collaboration brings together researchers and institutions around three research streams: 1. Nutrition and Physical Activity in chronic disease prevention: let by Professor Kerin O'Dea; 2. Mental Health and Substance Misuse in chronic disease prevention: Led by A/Professor Alan Clough; 3. Clinical Systems improvement in Primary Care: led by Professor Robyn McDermott.

The VecNet project supports a consortium of institutions to analyze malaria transmission and its reduction by vector and drug based interventions. The project will facilitate the development of new vector control tools, collection of new entomological data and analysis of transmission risk as a function of vector ecology and behaviour. It will also support an analysis of the composition of tools needed to achieve malaria eradication globally. Importantly, it will also provide input about the potential impact of candidate new tools, to inform prioritization of investments.

Tuberculosis is a major global public health problem and continues to cause significant morbidity and mortality due to the low efficiency of the only licensed anti-tuberculosis vaccine, BCG. This project aims to unravel the immunological correlates that mediate protection against tuberculosis. Using high-dimensional immune profiling and microbiome analyses we will compare immune responses to BCG vaccination with Mycobacterium tuberculosis infection in children in the Torres Strait.

This instrument is specifically designed for the rapid, robust, versatile, reproducible, and quantitative extraction of nucleic acids, proteins, and small molecules from a wide variety of sources including viruses, bacteria, plant and animal cells and tissues for mass spectrometry applications in genomics, proteomics, and metabolomics. This will drastically improve the reproducibility, speed and cost efficiency and reduces potential contaminations during sample preparation to generate very high quality data.

One of the major inequities in delivery of health services and of health outcomes occurs at a subnational level with more remote parts of countries suffering poorer health outcomes and lower levels of services. For many diseases, including TB, this is made more difficult to assess owing to poor surveillance infrastructure in remote communities. Hence there are an estimated 3 million missing cases of tuberculosis each year. This can be addressed by geospatial analysis and modelling to identify hidden hotspots and areas of need. Increasingly, more unified and complete data collection systems are in place (DHIS2) allowing geospatial analysis of data sources for facility-based and community-based activities. Understanding the health systems along which these operate; such as referral pathways, and patient/medication/service transport networks allows for simulation of new programs.
